Background Information

PANK4 (Pantothenate Kinase 4) is a member of the pantothenate kinase family, which is involved in the biosynthesis of coenzyme A (CoA). However, unlike other members of the family, PANK4 lacks the residues required for kinase activity and instead contains a phosphatase domain (DUF89) that can dephosphorylate 4'-phosphopantetheine, an intermediate in CoA biosynthesis (PMID: 30927326). This suggests that PANK4 may act as a pseudoenzyme, regulating cellular CoA levels by antagonizing CoA production (PMID: 39746949).
