Background Information

Zeta-associated protein (ZAP)-70, a member of the Syk family of tyrosine kinases, plays an important role in T-cell receptor signaling, natural killer (NK) cell activation and early B-cell development. Three tyrosine residues (Tyr292, Tyr315 and Tyr319) in the interdomain B region of ZAP‐70 have attracted particular attention as sites that might regulate ZAP‐70 activity and/or binding to heterologous proteins. The third candidate phosphorylation site in the interdomain B region of ZAP‐70 is Tyr319. Although the functional importance of this site in ZAP‐70 is unknown, a previous report presented indirect evidence that the orthologous Tyr352 residue in Syk is involved in the association of PLC‐γ1 with this PTK.
