  human placenta tissue were subjected to SDS PAGE followed by western blot with 83627-1-RR (Timp-3 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83627-1-PBS in a different storage buffer formulation.

  FC experiment of MDA-MB-231 using 83627-1-RR (same clone as 83627-1-PBS)

  1x10^6 MDA-MB-231 cells were intracellularly stained with 0.25 ug Timp-3 Recombinant antibody (83627-1-RR, Clone:240374D8) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer. This data was developed using the same antibody clone with 83627-1-PBS in a different storage buffer formulation.

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug Timp-3 Recombinant antibody (83627-1-RR, Clone:240374D8) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue).   Affinity and Kinetic Characterization of 83627-1-RR

  Biolayer interferometry (BLl) kinetic assays of 83627-1-RR against Human Timp-3 were performed. The affinity constant is 2.63 nM.

Background Information

Timp-3 (TIMP metallopeptidase inhibitor 3), also known as SFD. It is expected to be located in extracellular space, and the protein is enriched in the placenta tissue and fat tissue. The proteins encoded by this gene family are inhibitors of the matrix metalloproteinases, a group of peptidases involved in degradation of the extracellular matrix (ECM). Expression of this gene is induced in response to mitogenic stimulation, and this netrin domain-containing protein is localized to the ECM. Mutations in this gene have been associated with the autosomal dominant disorder Sorsby's fundus dystrophy. TIMP-3 is expressed as an unglycosylated 24 kDa and glycosylated 29 kDa protein with inhibitory activity against interstitial collagenase, stromelysin-1, and gelatinases A and B (PMID:11827795).
