Background
IGSF8 (CD316/EWI‑2) is a type I transmembrane glycoprotein of immunoglobulin superfamily, interacting tightly with tetraspanins CD9 and CD81 to modulate membrane trafficking and cell adhesion. Widely expressed on immune and neuronal cells, it functions as a novel innate immune checkpoint: overexpressed in diverse malignancies, it binds NK‑cell KIR3DL2 to block NK‑mediated tumor clearance. Preclinical and early clinical data validate it as a promising immunotherapy target; blocking anti‑IGSF8 mAbs boost antitumor immunity alone or combined with anti‑PD1 regimens.
