Background Information
ALG1 (asparagine-linked glycosylation 1) is a β-1,4-mannosyltransferase that catalyzes the committed step of N-linked glycosylation. Operating on the cytoplasmic face of the endoplasmic reticulum, ALG1 transfers the first mannose from GDP-mannose onto dolichol-pyrophosphate-GlcNAc₂, yielding Man₁GlcNAc₂-PP-dolichol. ALG1 is a 464-amino-acid protein with an N-terminal transmembrane domain that is essential for catalytic activity; deleting this domain abolishes mannosyltransferase function.
