1x10^6 HeLa cells were intracellularly stained with 0.25 ug FPGS Monoclonal antibody (60635-1-Ig, Clone:2D10H11) and CoraLite®488-Conjugated Goat Anti-Mouse IgG(H+L) (SA00013-1)(red), or 0.25 ug Mouse IgG1 isotype control Mouse McAb (66360-1-Ig, Clone: 1F8D3) (blue). Cells were fixed and permeabilized with Intracellular Flow Cytometry Fixation & Permeabilization Buffer Kit (PF00019). This data was developed using the same antibody clone with 60635-1-PBS in a different storage buffer formulation.

Background Information

FPGS is located in the cytosol and mitochondria of mammalian tissues, with current evidence supporting a single gene encoding for both the cytosolic and mitochondrial isoenzymes. FPGS catalyzes a rate-limiting polyglutamylation step in the folic acid metabolic pathway and increases the intracellular retention of folates (PMID: 8907263, PMID: 31611592). FPGS has 4 isoforms with the molecular mass of 59-65 kDa.
