  FC experiment of human peripheral blood leukocytes using 98086-1-RR (same clone as 98086-1-PBS)

  1x10^6 human peripheral blood leukocytes were surface stained with 0.25 ug Anti-Human CD177 Rabbit Recombinant Antibody (98086-1-RR, Clone:241339F4) or Isotype Control and PE-conjugated Goat Anti-Rabbit IgG. Cells were incubated with FC Receptor Block prior to fixing and staining. Cells were not fixed. This data was developed using the same antibody clone with 98086-1-PBS in a different storage buffer formulation.

  1x10^6 FC receptor blocked human peripheral blood leukocytes were surface stained with 0.25 ug Anti-Human CD177 Rabbit Recombinant Antibody (98086-1-RR, Clone:241339F4) and PE-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were not fixed. This data was developed using the same antibody clone with 98086-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 98086-1-RR

  Biolayer interferometry (BLl) kinetic assays of 98086-1-RR against Human CD177 were performed. The affinity constant is 1.70 nM.

Background Information

CD177, also known as NB1, HNA-2a or PRV-1, is a glycosylphosphatidylinositol (GPI)-anchored glycoprotein that belongs to the uPAR/CD59/Ly6 snake toxin superfamily (PMID: 17898576). It is expressed exclusively by neutrophils, neutrophilic metamyelocytes, and myelocytes (PMID: 17580308). CD177 interacts with β2 integrin CD11b/CD18 and CD11a/CD18, PECAM-1, and participates in neutrophil activation and transmigration (PMID: 17580308; 21193407). CD177 has been identified as a useful biomarker for myeloproliferative diseases (PMID: 17898576).
