Background Information

FAM175B (also known as ABRO1/KIAA0157) is a scaffold subunit of the BRCC36-containing deubiquitinase complex (BRISC) that specifically cleaves Lys63-linked polyubiquitin chains, thereby modulating DNA-damage responses and receptor trafficking. Originally identified as a binding partner of JunD and ATF4, FAM175B stabilizes ATF4 by inhibiting its ubiquitin-dependent degradation, leading to activation of the ATF4-CHOP pathway and induction of apoptosis under endoplasmic reticulum stress. In esophageal squamous cell carcinoma, FAM175B expression is down-regulated and acts as a p53-independent tumor suppressor; restoring its expression reduces proliferation and increases apoptosis of p53-mutated tumor cells. Collectively, FAM175B serves as a critical scaffold that links deubiquitinase activity to transcriptional and pro-apoptotic programs, offering a potential biomarker for early diagnosis and therapeutic targeting in cancers with p53 dysfunction (PMID: 30854784).
