KE00545 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human ALKBH3 ELISA kit is to be used to detect and quantify protein levels of endogenous human ALKBH3. The assay recognizes human ALKBH3. An antibody specific for human ALKBH3 has been pre-coated onto the microwells. The human ALKBH3 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human ALKBH3 is added to detect the captured human ALKBH3 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
ALKBH3, also named as ABH3 and DEPC-1, is a dioxygenase that mediates demethylation of DNA and RNA containing 1-methyladenosine (m1A). It has a strong preference for single-stranded. ALKBH3 is requires molecular oxygen, alpha-ketoglutarate and iron. This gene has some isoforms with MW 19-25 kDa and 33 kDa. Demethylates N1-methyladenosine (m1A) RNA, an epigenetic internal modification of messenger RNAs (mRNAs) highly enriched within 5'-untranslated regions (UTRs) and in the vicinity of start codons. Requires molecular oxygen, alpha-ketoglutarate and iron.
