KE00623 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human APCS/Serum amyloid P component ELISA kit is to be used to detect and quantify protein levels of endogenous human APCS/Serum amyloid P component. The assay recognizes human APCS/Serum amyloid P component. An antibody specific for human APCS/Serum amyloid P component has been pre-coated onto the microwells. The human APCS/Serum amyloid P component protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human APCS/Serum amyloid P component is added to detect the captured human APCS/Serum amyloid P component protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Serum amyloid P component (SAP), a member of pentraxin family, is a constitutive serum protein that is synthesized by hepatocytes. It is an acute phase protein, structurally related to C-reactive protein. SAP is a calcium-dependent ligand binding protein, which can interact with DNA and histones and may scavenge nuclear material released from damaged circulating cells.
