KE00486 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Aquaporin 4 ELISA kit is to be used to detect and quantify protein levels of endogenous human Aquaporin 4. The assay recognizes human Aquaporin 4. An antibody specific for human Aquaporin 4 has been pre-coated onto the microwells. The human Aquaporin 4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Aquaporin 4 is added to detect the captured human Aquaporin 4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Aquaporins are specialized water transport channels in plasma membranes of water-permeable tissues. Aquaporin-4 (AQP4) is the most abundant water channel in the human central nervous system and is important to fluid movements in brain. AQP4 is the principal water channel protein in the CNS, predominantly expressed in astrocyte endfeet and ependymal cells. It regulates water balance in brain tissue. Aquaporin-4 exists as two isoforms, a long (M1) isoform with translation initiation at Met-1, and a shorter (M23) isoform with translation initiation at Met-23, with molecular weights around 35-37 kDa and 32-34 kDa, respectively.
