KE00498 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Beta Arrestin 2 ELISA kit is to be used to detect and quantify protein levels of endogenous human Beta Arrestin 2. The assay recognizes human Beta Arrestin 2. An antibody specific for human Beta Arrestin 2 has been pre-coated onto the microwells. The human Beta Arrestin 2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Beta Arrestin 2 is added to detect the captured human Beta Arrestin 2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Beta - Arrestin 2 is a member of the arrestin family, which is involved in the regulation of G - protein - coupled receptor (GPCR) signaling. It is a cytosolic protein that is recruited to the plasma membrane upon GPCR activation. Beta - Arrestin 2 is involved in multiple antiproliferative pathways. It regulates the type III transforming growth factor beta (TGF - β) receptor and maternal embryonic leucine - zipper kinase (MELK), resulting in decreased TGF - β signaling and promoting the antiproliferative MELK pathway. eta - Arrestin 2 is involved in the pathogenesis of neurodegenerative diseases such as Parkinson's disease. It can affect the of accumulation alpha - synuclein and the function of dopaminergic neurons. Morphine tolerance may be mediated mainly via beta-arrestin 2.
