KE00434 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human CLEC12A ELISA kit is to be used to detect and quantify protein levels of endogenous human CLEC12A. The assay recognizes human CLEC12A. An antibody specific for human CLEC12A has been pre-coated onto the microwells. The human CLEC12A protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human CLEC12A is added to detect the captured human CLEC12A protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CLEC12A, also known as CD371, MICL, DCAL-2 or CLL-1, is a type II transmembrane glycoprotein and a member of the C-type lectin superfamily. This protein, which contains a single C-type lectin-like domain and a cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM), is related to LOX-1 and Dectin-1 and is variably spliced and highly N-glycosylated. Its ITIM sequence recruits the tyrosine phosphatases SHP-1 and SHP-2 to negatively regulate Syk signaling. In humans, CLEC12A is mainly expressed on myeloid cells, being found on monocytes, granulocytes, macrophages and DC subsets, and at lower levels on CD4+ T cells.
