KE00169 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The E-Selectin ELISA kit is to be used to detect and quantify protein levels of endogenous E-Selectin. The assay recognizes human E-Selectin. An antibody specific for E-Selectin has been pre-coated onto the microwells. The E-Selectin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for E-Selectin is added to detect the captured E-Selectin protein.For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm .

Background Information

E-selectin, also known as ELAM-1 or CD62E, is a member of the selectin family of adhesion molecules that also include L-selectin and P-selectin. E-selectin is an inducible endothelial cell surface molecule. Its expression on endothelial cells is transcriptionally upregulated by various proinflammatory substances such as IL-1, TNFα and lipopolysaccharide (LPS). Besides, it can be secreted by endothelial cells, and can be detected in a soluble form (sE-selectin) in serum. During inflammation, E-selectin plays an important part in recruiting leukocytes to the site of injury.
