Product Information

KE00265 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The HE4/WFDC2 ELISA kit is to be used to detect and quantify protein levels of endogenous human HE4/WFDC2. An antibody specific for HE4/WFDC2 has been pre-coated onto the microwells. The HE4/WFDC2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody specific for HE4/WFDC2 is added to detect the captured HE4/WFDC2 protein. For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Human epididymis protein 4 (HE4), also known as WFDC2, was originally identified as a transcript exclusively expressed in the epididymis and was proposed as a specific marker for this tissue. HE4 is a member of the Whey Acidic Protein (WAP) domain family of proteins. HE4 is expressed in a number of normal human tissues outside of the male reproductive system, including the trachea, lung and nasal epithelium and is also found in a subset of pulmonary epithelial-derived tumour cell lines. At present, HE4 serves as a serum biomarker of ovarian cancer, and has better sensitivity and specificity than CA125.
