KE00134 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human LBP ELISA kit is to be used to detect and quantify protein levels of endogenous human LBP. The assay recognizes human LBP. An antibody specific for human LBP has been pre-coated onto the microwells. The human LBP protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human LBP is added to detect the captured human LBP protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm .

Background Information
LBP (lipopolysaccharide-binding protein) is a serum glycoprotein belonging to the BPI/LBP/Plunc superfamily and BPI/LBP family. It is an acute-phase protein, predominantly synthesized by hepatocytes. LBP is present in normal serum at concentrations of 5 to 10 μg/ml, rising up to 200 μg/ml 24 h after induction of an acute-phase response. LBP binds to the lipid A moiety of bacterial lipopolysaccharides (LPS) which is a glycolipid present in the outer membrane of all Gram-negative bacteria, and acts as an affinity enhancer for CD14, facilitating its association with LPS.
