Product Information

KE00291 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human LIGHT/TNFSF14 ELISA kit is to be used to detect and quantify protein levels of endogenous human LIGHT/TNFSF14. The assay recognizes human LIGHT/TNFSF14. An antibody specific for human LIGHT/TNFSF14 has been pre-coated onto the microwells. The human LIGHT/TNFSF14 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human LIGHT/TNFSF14 is added to detect the captured human LIGHT/TNFSF14 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

LIGHT, also known as TNFSF14, HVEML, or CD258, is a type II transmembrane protein that belongs to the TNF superfamily. It is predominantly expressed in lymphoid tissues and produced by activated T cells and immature dendritic cells (DCs). Through its two primary functional receptors HVEM (TNFRSF14) and lymphotoxin β receptor (LTβR), LIGHT signaling is involved in development and maintenance of lymphoid tissues, as well as innate and adaptive immune responses. In addition to the membrane form, LIGHT can also exist as a soluble form generated by cleavage of the extracellular portion of the membrane form.
