KE00396 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Neurogranin ELISA kit is to be used to detect and quantify protein levels of endogenous human Neurogranin. The assay recognizes human Neurogranin. An antibody specific for human Neurogranin has been pre-coated onto the microwells. The human Neurogranin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Neurogranin is added to detect the captured human Neurogranin protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Neurogranin (formerly designated p17, also known as Ng, RC3 and BICKS) belongs to the neurogranin family. It is a neuron-specific substrate for protein kinase C (PKC). It is a postsynaptic protein that is highly enriched in brain, with restricted expression in the cortex, striatum, hippocampus, thalamus, hypothalamus and olfactory bulb nuclei. Neurogranin binds calmodulin at low levels of calcium, thereby regulating calmodulin-dependent nitric oxide synthase. Neurogranin acts as a "third messenger" substrate of protein kinase C-mediated molecular cascades during synaptic development and remodeling. It binds to calmodulin in the absence of calcium.
