KE00649 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Osteoprotegerin/TNFRSF11B ELISA kit is to be used to detect and quantify protein levels of endogenous human Osteoprotegerin/TNFRSF11B. The assay recognizes human Osteoprotegerin/TNFRSF11B. An antibody specific for human Osteoprotegerin/TNFRSF11B has been pre-coated onto the microwells. The human Osteoprotegerin/TNFRSF11B protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Osteoprotegerin/TNFRSF11B is added to detect the captured human Osteoprotegerin/TNFRSF11B protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Osteoprotegerin (OPG), also known as tumor necrosis factor receptor superfamily member 11B (TNFRSF11B) and osteoclastogenesis inhibitory factor (OCIF), belongs to the tumor necrosis factor (TNF) receptor superfamily. By binding to RANKL, OPG inhibits osteoclastic activity and promoting bone formationis. OPG plays a key role on cell survival by inhibiting TRAIL-induced apoptosis. OPG also is involved in vascular biology, fibrosis, immunity, EMT, and the apoptosis of cancer cells.
