KE00842 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human NCX1 ELISA kit is to be used to detect and quantify protein levels of endogenous human NCX1. The assay recognizes human NCX1. An antibody specific for human NCX1 has been pre-coated onto the microwells. The human NCX1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human NCX1 is added to detect the captured human NCX1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
RAF1, also known as Raf-1 proto-oncogene serine/threonine kinase, plays pivotal roles in the MAPK/ERK signaling pathway. It is activated by growth factor receptors and other extracellular stimuli, promoting cell proliferation, differentiation, and survival. Upon activation, RAF1 phosphorylates and activates MEK, which in turn activates ERK. Abnormal activation of RAF1 is implicated in various cancers. Mutations or overexpression of RAF1 can lead to constitutive activation of the MAPK pathway, driving cell proliferation and tumorigenesis. For example, RAF1 is involved in the development of multiple tumors, such as non-small cell lung cancer.
