KE00614 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human/rat NEDD4 ELISA kit is to be used to detect and quantify protein levels of endogenous human/rat NEDD4. The assay recognizes human/rat NEDD4. An antibody specific for human/rat NEDD4 has been pre-coated onto the microwells. The human/rat NEDD4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human/rat NEDD4 is added to detect the captured human/rat NEDD4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
NEDD4 and similar proteins discovered subsequently became a family of HECT ligases, comprising 9 human proteins including NEDD4, NEDD4-2 (NEDD4L), ITCH, SMURF1, SMURF2, WWP1, WWP2, NEDL1 and NEDL2. NEDD4 is a highly evolutionarily conserved protein from yeast to man, and was initially cloned as a highly expressed gene in the early embryonic brain. NEDD4 is frequently overexpressed in many different types of cancer, and decreased levels of NEDD4 can also be associated with cancer. It can be a potential therapeutic target for the treatment of human cancer.
