KE00851 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human SAMHD1 ELISA kit is to be used to detect and quantify protein levels of endogenous human SAMHD1. The assay recognizes human SAMHD1. An antibody specific for human SAMHD1 has been pre-coated onto the microwells. The human SAMHD1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human SAMHD1 is added to detect the captured human SAMHD1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
SAMHD1, highly expressed by monocytes and monocyte-derived dendritic cells and by monocyte-derived macrophages at a lower extent, is reported as an HIV-1 restriction factor that inhibits the early step of the HIV-1 life cycle. Vpx (virus-like particles containing viral protein X) could overcome this block by interacting with SAMHD1, inducing proteasome-dependent degradation of SAMHD1. Mutations in SAMHD1 cause Aicardi-Goutières syndrome, a genetic encephalopathy with presumed immune pathogenesis.
