KE00806 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human SMARCA2 ELISA kit is to be used to detect and quantify protein levels of endogenous human SMARCA2. The assay recognizes human SMARCA2. An antibody specific for human SMARCA2 has been pre-coated onto the microwells. The human SMARCA2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human SMARCA2 is added to detect the captured human SMARCA2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
SMARCA2 (also known as BRM) is an important chromatin remodeling protein and a core subunit of the SWI/SNF complex. It alters chromatin structure through its ATPase activity to regulate gene expression and is involved in various biological processes, including cell differentiation, proliferation, and DNA damage repair. SMARCA2 and SMARCA4 (BRG1) are mutually exclusive in different SWI/SNF complexes and work synergistically in the regulation of gene expression. Dysfunction of SMARCA2 is associated with multiple diseases, including various cancers (such as pancreatic cancer, lung cancer, and ovarian cancer) and genetic disorders (such as Nicolaides-Baraitser syndrome).
