KE00834 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human cIAP2 ELISA kit is to be used to detect and quantify protein levels of endogenous human cIAP2. The assay recognizes human cIAP2. An antibody specific for human cIAP2 has been pre-coated onto the microwells. The human cIAP2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human cIAP2 is added to detect the captured human cIAP2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Cellular inhibitor of apoptosis protein 2 (cIAP2) is also named as baculoviral IAP repeat-containing protein 3 (BIRC3), hIAP-1, MIHC, RNF49 and Apoptosis inhibitor 2 (API2). cIAP2 is an essential member of IAPs family. Its combination with receptor-interacting protein kinase 1 (RIPK1) could activate NF-κb and MAPK pathways for cell survival. cIAP2 is a RING-containing protein ubiquitin ligase. cIAP2 normally inhibits antigen receptor signaling by mediating the ubiquitination and degradation of Bcl10, a critical component for antigenic signaling to NF-kappaB. cIAP2 as being highly expressed in SOX10-deficient drug tolerant persister (DTP) melanoma cells. cIAP2 is up-regulated in SOX10-deficient cells at the transcript level by the AP-1 protein complex component JUND. cIAP2 expressing cells can be targeted with the IAP inhibitor, birinapant, through induction of cell death. Many cancers express high levels of cIAP2 and other IAPs. For example, all four IAP family members were commonly elevated in prostate cancers, and cIAP2 expression was elevated in human colon cancer cells.

FAQ
1、I have limited sample diluent. How should I handle samples requiring high dilution factors?
Answer: We recommend performing a serial dilution. For example, if human serum requires a 1,000,000-fold dilution, you can use a three-step 100× dilution method as follows:
Step 1: Take 2 uL of human serum and add it to 198 uL of sample diluent, mix thoroughly to obtain a 100-fold diluted sample.
Step 2: Take 2 uL of the 100-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 10,000-fold diluted sample.
Step 3: Take 2 uL of the 10,000-fold diluted sample and add it to 198 uL of sample diluent, mix thoroughly to obtain a 1,000,000-fold diluted sample.
If further dilution is required, additional serial steps can be used. We do not recommend using any other solutions or water for dilution, except for the sample diluent we suggest. If your sample diluent is still insufficient, please contact our customer service for more sample diluent.
