  WB analysis of K-562 using 83471-5-RR (same clone as 83471-5-PBS)

  K-562 cells were subjected to SDS PAGE followed by western blot with 83471-5-RR (IKZF4 antibody) at dilution of 1:20000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83471-5-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (4% PFA) fixed paraffin-embedded mouse stomach tissue using IKZF4 antibody (83471-5-RR, Clone: 240357D5 ) at dilution of 1:200 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 83471-5-PBS in a different storage buffer formulation.

  FC experiment of U2OS using 83471-5-RR (same clone as 83471-5-PBS)

  1x10^6 U2OS cells were intracellularly stained with 0.25 ug IKZF4 Recombinant antibody (83471-5-RR, Clone:240357D5) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 83471-5-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83471-5-RR

  Biolayer interferometry (BLl) kinetic assays of 83471-5-RR against Human IKZF4 were performed. The affinity constant is 21.3 nM.

Background Information

IKZF4 is a member of the Ikaros family transcription factors. The Ikaros family was originally designated to function in hematopoiesis, but it is only recently that studies have begun to show the relevance of this family in T helper cell differentiation. IKZF3 was preferentially expressed in TH17 cells and governs TH17 differentiation by silencing IL-2 expression. By investigating the dynamics of the TH17 regulatory network, IKZF4 was proposed by Yosef et al. as a composite in the negative regulatory module for TH17. (PMID: 24644282)
