Background Information

KCC4, also named as Solute carrier family 12 member 7(SLC12A7), is a 1083 amino-acid protein, which belongs to the SLC12A transporter family. As a multi-pass membrane protein, the molecular weight of KCC4 is 119 kDa. KCC4 mediates electroneutral potassium-chloride cotransport when activated by cell swelling. Moreover, KCC4 may mediate K+ uptake into Deiters' cells in the cochlea and contribute to K+ recycling in the inner ear. KCC4 is important for the survival of cochlear outer and inner hair cells and the maintenance of the organ of Corti(Uniprot, GeneID:10723).

In the Western blots there is an additional band below at 98kD which seems like a degradation or non-specific binding of the antibody. The bands showing at 120-130kD are accurate as we have seen in the lanes from knockdowns and overexpression of protein KCC4 For reference information about the lanes: 1. MDA KCC4 overexpression 2. MDA KCC4 knockdown sequence1 3. MDA KCC4 knockdown sequence2 4. MDA KCC4 knockdown sequence3 5. MDA scrambled control As you can see in lane 5 which is the control has expression of kCC4 at 120-130 range and for the knockdowns there is a decrease more significantly for 2nd sequence and for overexpression in lane 1 it is quite high so we believe the top band is the expected one and the bottom one is a result of degradation maybe so please advice on this matter. As for the antibody we received it in April 2023 and it is stored at -20 as mentioned.
