Background Information

MAF1 operates as a conserved repressor of RNA polymerase III transcription, first identified in yeast. It binds RNA pol III directly, blocking TFIIIB recruitment and dampening tRNA and 5S rRNA output. In mammals, MAF1 additionally fine-tunes select RNA pol II genes, impacting metabolism, bone formation, adipogenesis, and oncogenesis, while its own activity is stringently controlled by mTORC1 phosphorylation.
