Product Information

KE10177 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse CTLA-4 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse CTLA-4. The assay recognizes mouse CTLA-4. An antibody specific for mouse CTLA-4 has been pre-coated onto the microwells. The mouse CTLA-4 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse CTLA-4 is added to detect the captured mouse CTLA-4 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4) is a co-inhibitory receptor that plays a crucial role in regulating T-cell activation and maintaining immune tolerance and delivers inhibitory signals that suppress T-cell responses, thereby preventing autoimmune diseases. It is expressed on activated T cells and regulatory T cells (Tregs), which mediates its inhibitory function by competing with CD28 for binding to CD80/CD86, depleting these ligands from antigen-presenting cells (APCs) and dendritic cells (DCs), and delivering inhibitory signals via its intracellular tail. CTLA-4 exists in two forms: membrane-bound CTLA-4 (mCTLA-4) and soluble CTLA-4 (sCTLA-4). mCTLA-4 is a transmembrane protein localized on the surface of T cells and Tregs, where it interacts with CD80 and CD86 on APCs. sCTLA-4 lacks the transmembrane domain and is released into the extracellular environment, where it can modulate immune responses.
