Product Information
KE10049 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The CXCL12 ELISA kit is to be used to detect and quantify protein levels of endogenous CXCL12. The assay recognizes mouse CXCL12. An antibody specific for CXCL12 has been pre-coated onto the microwells. The CXCL12 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for mouse CXCL12 is added to detect the captured mouse CXCL12 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
The chemokine stroma-derived factor (SDF-1/CXCL12) plays multiple roles in tumor pathogenesis. CXCL12 plays an important role in angiogenesis by recruiting endothelial progenitor cells (EPCs) from the bone marrow through a CXCR4 dependent mechanism (PMID: 17878755). It is this function of CXCL12 that makes it a very important factor in carcinogenesis and the neovascularisation linked to tumour progression (PMID: 16943240). CXCL12 also has a role in tumor metastasis where cancer cells that express the receptor CXCR4 are attracted to metastasis target tissues that release the ligand, CXCL12 (PMID: 11242036 ). In breast cancer, however, increased expression of CXCL12 determines a reduced risk of distant metastasis (PMID: 19646861 ).
