KE10164 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse GSDMD ELISA kit is to be used to detect and quantify protein levels of endogenous mouse GSDMD. The assay recognizes mouse GSDMD. An antibody specific for mouse GSDMD has been pre-coated onto the microwells. The mouse GSDMD protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse GSDMD is added to detect the captured mouse GSDMD protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Gasdermin D (GSDMD) is a member of the gasdermin family and a key pore-forming protein that plays a crucial role in regulating inflammatory responses and various forms of cell death, particularly pyroptosis. GSDMD is activated by inflammatory caspases, such as caspase-1, -4, -5, and -11, which cleave it into an N-terminal fragment (GSDMD-N) and a C-terminal fragment (GSDMD-C). GSDMD has both pro-inflammatory and protective roles depending on the context. In some infectious diseases and inflammatory conditions, GSDMD helps in pathogen clearance and immune defense. In tumor diseases, GSDMD exhibits dual regulatory roles: it can suppress tumor growth but may also promote tumor cell migration and metastasis under certain conditions. Targeting GSDMD has emerged as a promising therapeutic strategy for various inflammatory and non-tumor diseases
