KE10084 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse PIGF-2 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse PIGF-2.The assay recognizes mouse PIGF-2. An antibody specific for mouse PIGF-2 has been pre-coated onto the microwells. The mouse PIGF-2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for mouse PIGF-2 is added to detect the captured mouse PIGF-2 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Placenta growth factor (PlGF, also called PGF) is a member of the PDGF/VEGF family of secreted growth factors that share a conserved pattern of eight cysteines. PlGF is a homodimeric glycoprotein encoded by a single PlGF gene that gives rise to four splice isoforms in humans, PlGF-1 (PlGF131), PlGF-2 (PlGF152), PlGF-3 (PlGF203) and PlGF-4 (PlGF224). PlGF-1 and PlGF-2 represent the predominant isoforms. Unlike VEGF, PlGF homodimers bind VEGFR1, but not VEGFR2. PlGF exerts different non-redundant effects in stress conditions including wound healing, tissue ischemia and sepsis. PlGF-2 is expressed in human umbilical vein endothelial cells and in placenta.
