  Cytometric bead array of cell line supernatant using MP51307-1, BMP-4 Monoclonal Matched Antibody Pair, PBS Only. Capture antibody: 60897-1-PBS. Detection antibody: 69015-1-PBS. Supernatant (50μL/well) from the following cell lines were assayed: HT-29, TF-1.

  Neutralization experiment of NeutraKine® BMP-4 using 69015-1-Ig (same clone as 69015-1-PBS)

  Recombinant human BMP-4 (Cat.NO. HZ-1045) induces alkaline phosphatase production in the ATDC-5 cell line (using pNPP as chromogenic substrate for detection) in a dose dependent manner (blue curve, bottom X-left Y). The activity of human BMP-4 (200 ng/mL HZ-1045) is neutralized by mouse anti-human BMP-4 antibody 69015-1-Ig at serial dose (red curve, refer to top X-right Y). The ND50 is typically 3-10 μg/mL. This data was developed using the same antibody clone with 69015-1-PBS in a different storage buffer formulation.

Background Information

BMP4, also named as BMP2B and DVR4, belongs to the TGF-beta family. BMPs have been shown to promote astroglial differentiation both in vitro and in vivo. BMP4 have been shown to be components of serum and are thought to be responsible for this differentiation. BMP4 are expressed mainly in neurons, with minimal expression detected in astrocytes and oligodendrocytes. This antibody can be used to neutralize the bioactivity of BMP-4.
