  Neutralization experiment of NeutraKine® Cystatin C using 69017-1-Ig (same clone as 69017-1-PBS)

  Recombinant human Cystatin C (Cat.NO. HZ-1211) inhibits Papain protease (5 μg/mL) activity (by colorimetric assay using L-BAPA as substrate) in a dose-dependent manner (blue curve, refer to bottom X-left Y). The activity of human Cystatin C (0.4 μg/mL HZ-1211) is neutralized by mouse anti-human Cystatin C monoclonal antibody 69017-1-Ig at serial dose (red curve, refer to top X-right Y). The ND50 is around 0.5mg/mL. This data was developed using the same antibody clone with 69017-1-PBS in a different storage buffer formulation.

Background Information

Cystatin C is a 13-kDa inhibitor of cysteine proteinases which is secreted by all cell types and is completely cleared from the organism through glomerular filtration, shown to be an early and sensitive biomarker of renal dysfunction. It is also used as an emerging biomarker in cardiovascular disease. Cystatin C is involved in a variety of inflammatory reactions. The concentration of serum cystatin C has also been shown to be unaltered in certain inflammatory conditions or other disorders of metabolism. The plasma level of serum cystatin C can be expressed as its level of generation from cells and diet and its subsequent elimination through the gut, liver, and kidneys.

This antibody can be used to neutralize the bioactivity of Cystatin C.
