Background Information
Prolyl-tRNA synthetase 2, mitochondrial (PARS2) is a nuclear-encoded mitochondrial aminoacyl-tRNA synthetase (mt-aaRS) responsible for catalyzing the attachment of proline to mitochondrial tRNAPro, an essential step in mitochondrial protein translation. Following cytoplasmic synthesis, PARS2 is imported into the mitochondrial matrix, where it ensures the accurate synthesis of the 13 oxidative phosphorylation (OXPHOS) proteins encoded by mitochondrial DNA. By maintaining the fidelity of mitochondrial translation, PARS2 is indispensable for mitochondrial respiratory chain function, ATP production, and cellular energy metabolism.
