Background Information
Protein O-fucosyltransferase 1 (POFUT1) is an endoplasmic reticulum-resident glycosyltransferase that transfers O-fucose from GDP-fucose to conserved serine or threonine residues within epidermal growth factor-like (EGF) repeats of cell surface and secreted proteins, most notably the Notch receptor family. This modification is essential for Notch signaling: POFUT1-mediated O-fucosylation on Notch EGF repeats directly modulates ligand binding and is required for proper receptor trafficking and canonical signal activation, while also serving as a prerequisite for further glycan elongation by Fringe glycosyltransferases. Beyond Notch, POFUT1 fucosylates other EGF-repeat-containing substrates and functions in an endoplasmic reticulum quality control capacity, stabilizing folded EGF domains. Genetic ablation of Pofut1 in mice is embryonic lethal with phenotypes mirroring global Notch inactivation, and heterozygous mutations in human POFUT1 have been linked to Dowling-Degos disease, underscoring its physiological importance in development and tissue homeostasis.

Reviews
What customers say
The single review for this antibody reports a 4-star rating from use in Western Blot, Immunoprecipitation, and Immunofluorescence on pancreatic beta cells. The reviewer found the overall results to be good across all three applications, noting that the weaker signal observed in Western Blot may be attributable to the specific cell type used rather than the antibody itself. No dilution details were provided.
The reviews below have been submitted by verified Proteintech customers who received an incentive for providing their feedback.
fei (Verified Customer) (09-04-2025) | This antibody was used for WB, IP, and IF experiments. Overall, the results were good. The weaker signal in WB may be related to the cell type.
