Background Information
PTOV1 is a dual-domain adaptor and transcriptional co-regulator that promotes cell proliferation and oncogenic signaling through interactions with E2F1, RSK, and Mediator components. Shuttling between cytoplasm and nucleus, it integrates growth factor and differentiation pathways to balance proliferation and differentiation. Overexpressed in multiple cancers-most prominently prostate carcinoma-PTOV1 is linked to tumor progression, therapy resistance, and poor prognosis, positioning it as a potential biomarker and therapeutic target in epithelial malignancies.
