Background Information

CAD (carbamoyl-phosphate synthetase 2, aspartate transcarbamoylase, and dihydroorotase) is a multifunctional enzyme required for the de novo synthesis of pyrimidine nucleotides. Phospho-CAD (Ser1859) refers to the phosphorylated form of the multi-enzyme CAD (carbamoyl-phosphate synthetase 2, aspartate transcarbamylase, and dihydroorotase) on serine 1859. This modification is catalyzed by ribosomal protein S6 kinase 1 (S6K1) downstream of mTORC1 signaling and serves as a metabolic switch that converts CAD from its basal state into a highly active oligomer, thereby accelerating the first three rate-limiting steps of de novo pyrimidine synthesis.
