Background Information

HER2, also known as ErbB2 and Neu, is a 185-kDa transmembrane glycoprotein that is a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. Phospho-HER2/ErbB2 (Tyr1196) is a phosphorylated form of the HER2 protein, which is a receptor tyrosine kinase that plays a critical role in cell growth, proliferation, and survival. The phosphorylation of HER2 at Tyr1196 is associated with the activation of downstream signaling pathways. In breast cancer, HER2 is overexpressed and contributes to aggressive tumor growth and poor prognosis. Overexpression of Her2/ErbB2/Neu in cancer is often correlated with recurrent distant metastasis, although the mechanism still remains largely elusive. Tyr1139/Tyr1196 are autonomously phosphorylated, when Her2 is overexpressed. (PMID: 21966491)
