  WB analysis of HT-29 using 84403-1-RR (same clone as 84403-1-PBS)

  Non-treated HT-29 cells and TNF alpha and Calyculin A treated HT-29 cells were subjected to SDS PAGE followed by western blot with 84403-1-RR (Phospho-IkB Beta (Thr19/Ser23) antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Alpha Tubulin (66031-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 84403-1-PBS in a different storage buffer formulation.

  FC experiment of HT-29 using 84403-1-RR (same clone as 84403-1-PBS)

  1x10^6 TNF alpha and Calyculin A treated or untreated(black) HT-29 cells were intracellularly stained with 0.25 ug Phospho-IkB Beta (Thr19/Ser23) Recombinant antibody (84403-1-RR, Clone:241061B2) and PE-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with 90% MeOH. This data was developed using the same antibody clone with 84403-1-PBS in a different storage buffer formulation.

Background Information

IkB Beta is also named as NF-kappa-B inhibitor beta (NFKBIB) and TRIP-9. IKBB is one of the major isoforms of the NFKB inhibitor which preferentially interacts with c-Rel or p65 NF-kB dimers (PMID: 26227166). IKBB was reported to be associated with tumor growth inhibition and metastasis in lung cancer (PMID: 11402320). IKBB also inhibits cancer cell regulation through inactivation of the NFkB pathway (PMID: 18950638). Exposure of cells to a variety of extracellular stimuli leads to the rapid phosphorylation, ubiquitination, and ultimately proteolytic degradation of IkB, which frees NF-kB to translocate to the nucleus where it regulates gene transcription (PMID: 10837071). The antibody is phosphorylated at Thr19 and Ser23.
