  WB analysis using 80077-9-RR (same clone as 80077-9-PBS)

  Non-treated HT-29 cells and etoposide treated HT-29 cells were subjected to SDS PAGE followed by western blot with 80077-9-RR (Phospho-P53 (Ser46) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with GAPDH (60004-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 80077-9-PBS in a different storage buffer formulation.

  Non-treated A431 cells and nocodazole treated A431 cells were subjected to SDS PAGE followed by western blot with 80077-9-RR (Phospho-P53 (Ser46) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with GAPDH (60004-1-Ig) antibody as a loading control.   IF Staining of HT-29 using 80077-9-RR (same clone as 80077-9-PBS)

  Immunofluorescent analysis of (4% PFA) fixed etoposide treated HT-29 cells using Phospho-P53 (Ser46) antibody (80077-9-RR, Clone: 250307A3 ) at dilution of 1:500 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2). This data was developed using the same antibody clone with 80077-9-PBS in a different storage buffer formulation.

Background Information

The p53 tumor suppressor protein plays a major role in cellular response to DNA damage and other genomic aberrations. Activation of p53 can lead to either cell cycle arrest and DNA repair or apoptosis. Phosphorylation of p53 at Ser46 regulates the ability of p53 to induce apoptosis. Homeodomain-interacting protein kinase 2 (HIPK2) Phosphorylates Ser46 off p53 and p38 can phosphorylate this site.
