WB analysis using 87019-1-RR (same clone as 87019-1-PBS)
Non-treated mouse brain tissue, phosphatase inhibitor treated mouse brain tissue and λ phosphatase treated mouse brain tissue were subjected to SDS PAGE followed by western blot with 87019-1-RR (Phospho-TAU(Ser409) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with GAPDH (60004-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 87019-1-PBS in a different storage buffer formulation.

Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 87019-1-PBS in a different storage buffer formulation.

Background Information

Phosphorylation of tau at Ser409 is an early, disease-specific event in Alzheimer's brain. Catalyzed by PKA, this modification weakens tau-microtubule binding and acts as a molecular switch that initiates tau self-aggregation into paired helical filaments. Consequently, p-Ser409-tau is recognized as an early diagnostic epitope and a potential target for interrupting PKA-driven, oxidative-stress-linked tau pathology.
