  WB analysis of HEK-293 using 80706-3-RR

  Non-treated HEK-293 cells, phosphatase inhibitor treated HEK-293 cells and λ phosphatase treated HEK-293 cells were subjected to SDS PAGE followed by western blot with 80706-3-RR (Phospho-c-SRC (Ser17) antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Beta Actin (66009-1-Ig) antibody as a loading control.

  FC experiment of HEK-293 using 80706-3-RR

  1X10^6 HEK-293 cells untreated (dashed lines) or treated with λ phosphatase which intracellularly stained with 0.13 ug Phospho-c-SRC (Ser17) Recombinant monoclonal antibody (80706-3-RR, Clone:250845F12) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.13 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 90% MeOH .

Background Information

SRC, also named as SRC1 and p60-Src, belongs to the protein kinase superfamily, Tyr protein kinase family and SRC subfamily. The phosphorylation of c-SRC at Ser17 is essential for its activation. This phosphorylation event is often mediated by protein kinase A (PKA) and is crucial for downstream signaling events. (PMID: 23360994)
