Background Information
TSSC1 (Tumor Suppressing Subtransferable Candidate 1), now officially designated EIPR1, encodes a 387-amino-acid WD40 repeat protein that localizes to chromosome 2p25.3 in humans. Although originally identified within a putative tumor-suppressor region at 11p15.5, later genomic mapping reassigned the gene to chromosome 2. Functionally, TSSC1 is a component of the endosomal retrieval machinery, interacting with both the EARP and GARP tethering complexes to regulate protein transport from early endosomes to the trans-Golgi network and recycling endosomes. Gershlick et al. demonstrated that TSSC1 is required for efficient recruitment of GARP to the trans-Golgi network and functions as a regulator of both GARP and EARP in endosomal retrieval pathways (PMID: 27440922).
