Background Information
MPIG6B (Megakaryocyte and Platelet Inhibitory Receptor G6b), also known as G6b-B or C6orf25, is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, and serves as a key negative regulatory receptor for platelet activation and megakaryocyte differentiation. The intracellular domain of MPIG6B contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs), which recruit SHP-1/SHP-2 phosphatases to inhibit Src family kinase activity, thereby negatively regulating platelet aggregation, release reactions, and megakaryocyte maturation. Mutations in the MPIG6B gene lead to thrombocytopenia and bleeding tendency, and its functional dysregulation is associated with immune thrombocytopenia (ITP) and myeloproliferative neoplasms.
