FC experiment of human PBMCs using 98674-2-RR
1x10^6 human PBMCs were surface stained with 0.25 ug Anti-Human ADAM17 Rabbit RecAb (98674-2-RR, Clone:251631A5), or Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Cells were then stained with APC Anti-Human CD14. Cells were incubated with FC Receptor Block prior to staining. Cells were not fixed.

Background Information

The ADAMs (A Disintegrin And Metalloprotease) are multidomain transmembrane proteins. One of the first ADAMs implicated in membrane shedding is ADAM-17, which is shown to release the active form of tumor necrosis factor (TNF)-a from its precursor (PMID:18238782). ADAM17 mediates the release of pro-inflammatory cytokines and modulates cell signaling pathways involved in tumor progression and metastasis (PMID:17438092).
