FC experiment of human PBMCs using 98674-2-RR (same clone as 98674-2-PBS)
1x10^6 human PBMCs were surface stained with 0.25 ug Anti-Human ADAM17 Rabbit RecAb (98674-2-RR, Clone:251631A5), or Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Cells were then stained with APC Anti-Human CD14. Cells were incubated with FC Receptor Block prior to staining. Cells were not fixed. This data was developed using the same antibody clone with 98674-2-PBS in a different storage buffer formulation.

Background Information

The ADAMs (A Disintegrin And Metalloprotease) are multidomain transmembrane proteins. One of the first ADAMs implicated in membrane shedding is ADAM-17, which is shown to release the active form of tumor necrosis factor (TNF)-a from its precursor (PMID:18238782). ADAM17 mediates the release of pro-inflammatory cytokines and modulates cell signaling pathways involved in tumor progression and metastasis (PMID:17438092).
