Background Information
AVPR2 is a Gs-coupled vasopressin receptor essential for renal water reabsorption, acting through cAMP-PKA-dependent trafficking of aquaporin-2. Localized to principal cells of the collecting duct, AVPR2 integrates hormonal cues to maintain osmotic and fluid homeostasis. Mutations lead to X-linked nephrogenic diabetes insipidus, while rare activating variants cause SIADH. Because of its central role in renal physiology and clear clinical associations, AVPR2 is a key target in water-balance disorders and pharmacologic modulation of diuresis.
