  WB analysis using 86644-1-RR (same clone as 86644-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 86644-1-RR (ALIX; AIP1 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 86644-1-PBS in a different storage buffer formulation.

  IF Staining of HeLa using 86644-1-RR (same clone as 86644-1-PBS)

  Immunofluorescent analysis of (-20°C Ethanol) fixed HeLa cells using Alix antibody (86644-1-RR, Clone: 251574C12 ) at dilution of 1:200 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2). This data was developed using the same antibody clone with 86644-1-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 86644-1-RR (same clone as 86644-1-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug Alix Recombinant monoclonal antibody (86644-1-RR, Clone:251574C12) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Intracellular Flow Cytometry Fixation & Permeabilization Buffer Kit (PF00019). This data was developed using the same antibody clone with 86644-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 86644-1-RR

  Biolayer interferometry (BLl) kinetic assays of 86644-1-RR against Human Alix were performed. The affinity constant is 6.12 nM.

Background Information

ALG-2-interacting protein 1 (ALIX), also known as AIP1 or Hp95, is encoded by PDCD6IP gene and is involved in cell death through mechanisms involving its binding partner ALG-2 (apoptosis-linked gene-2). ALG-2 is a 22-kDa protein containing five serially repetitive EF-hand structures and is defined as a regulator of calcium-induced apoptosis following endoplasmic reticulum (ER) stress. ALIX interacts with ALG-2 through its C-terminal proline-rich region and participates in formation of multivesicular bodies. Recent finding suggest that ALIX is a critical component of caspase 9 activation and apoptosis triggered by calcium. The alix antibody recognizes an additional band of 75-80 kDa which has also been observed in cells and exosomes.
