Product Information

KE00267 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Thrombopoietin(TPO) ELISA kit is to be used to detect and quantify protein levels of endogenous TPO. The assay recognizes human TPO. An antibody specific for human TPO has been pre-coated onto the microwells. The human TPO protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TPO is added to detect the captured human TPO protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Thrombopoietin (TPO) also known as megakaryocyte growth and development factor (MGDF), is a protein in human. The protein is a glycoprotein hormone produced by the liver and kidney, regulating the differentiation of megakaryocytes and platelets. While platelet and megakaryocyte concentrations have a negative feedback on TPO levels. It is also a humoral growth factor necessary for megakaryocyte proliferation and maturation, thrombopoiesis. TPO is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene and one of the Class I hematopoietic cytokines. TPO promotes apoptosis of newly generated neurons in brain, an effect counteracted by Erythropoietin and neurotrophins.
