  FC experiment of human peripheral blood leukocytes using 98293-1-RR (same clone as 98293-1-PBS)

  1x10^6 human peripheral blood leukocytes were surface stained with 0.25 ug Anti-Human CD16a Rabbit RecAb (98293-1-RR, Clone:242426G11) , or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) and PE-Conjugated Goat Anti-Rabbit IgG(H+L). Cells were incubated with FC Receptor Block prior to staining. Cells were not fixed. This data was developed using the same antibody clone with 98293-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 98293-1-RR

  Biolayer interferometry (BLl) kinetic assays of 98293-1-RR against Human CD16a were performed. The affinity constant is 14.3 nM.

  98293-1-RR specifically recognizes human CD16a

  Biolayer interferometry (BLl) kinetic assays of 98293-1-RR against Human CD16a 176F (A), CD16a 176V (B), CD16b NA1 (C) and CD16b NA2 (D) were performed. Affinity constants against two CD16a allotypes are 25.9 nM and 18.3 nM. No binding signal against CD16b allotypes was detected.

Background Information

CD16 is a 50-70-kDa low affinity Fc receptor found on the surface of natural killer cells, neutrophil polymorphonuclear leukocytes, monocytes and macrophages. CD16 mediates antibody-dependent cellular cytotoxicity (ADCC) and other antibody-dependent responses, such as phagocytosis. CD16 has been identified as Fc receptors FcγRIIIa (CD16a) and FcγRIIIb (CD16b), encoded by two nearly identical genes, FCGR3A and FCGR3B.
